Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt FH protein function. ClinVar contains an entry for this variant (Variation ID: 529807). This missense change has been observed in individuals with leiomyomas (Invitae). This variant is not present in population databases (gnomAD no frequency). -

The p.G326E variant (also known as c.977G>A), located in coding exon 7 of the FH gene, results from a G to A substitution at nucleotide position 977. The glycine at codon 326 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This alteration has been observed in at least one individual who has a personal or family history that is consistent with FH-associated disease (Ambry internal data; Franke K et al. Clin Case Rep, 2022 Mar;10:e05513). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic. -
