This variant is denoted PTEN c.740T>G at the cDNA level and p.Leu247Ter (L247X) at the protein level. The substitution creates a nonsense variant, which changes a Leucine to a premature stop codon (TTA>TGA), and is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. This variant has not, to our knowledge, been published in the literature as either a pathogenic germline variant or a benign polymorphism. However, it has been reported as a somatic variant in a melanoma cell line (Aguissa-Tour? 2012). We consider this variant to be pathogenic. -

The p.L247* pathogenic mutation (also known as c.740T>G) located in coding exon 7 of the PTEN gene, results from a T to G substitution at nucleotide position 740. This changes the amino acid from a leucine to a stop codon within coding exon 7. Since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). -
