In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available"). This variant has not been reported in the literature in individuals affected with PTEN-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces glutamic acid, which is acidic and polar, with aspartic acid, which is acidic and polar, at codon 285 of the PTEN protein (p.Glu285Asp).
Hereditary cancer-predisposing syndrome Uncertain:1
The p.E285D variant (also known as c.855A>T), located in coding exon 8 of the PTEN gene, results from an A to T substitution at nucleotide position 855. The glutamic acid at codon 285 is replaced by aspartic acid, an amino acid with highly similar properties. In a humanized yeast model, lipid phosphatase activity for this variant is hypomorphic (Mighell TL et al. Am. J. Hum. Genet., 2018 05;102:943-955). This variant demonstrated possibly wildtype-like intracellular protein abundance on one multiplex functional assay (Matreyek KA et al. Nat Genet, 2018 06;50:874-882). This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
