This protein is an important cell cycle checkpoint kinase that phosphorylates; thus, it functions as a regulator of a wide variety of downstream proteins, including tumor suppressor proteins p53 and BRCA1, checkpoint kinase CHK2, checkpoint proteins RAD17 and RAD9, and DNA repair protein NBS1. This protein and the closely related kinase ATR are thought to be master controllers of cell cycle checkpoint signaling pathways that are required for cell response to DNA damage and for genome stability. The threonine at codon 2921 is replaced by methionine, an amino acid with similar properties. This alteration has been previously identified in a compound heterozygous state with ATM p.C353R (c.1057 T>C) in 2 patients from a Saudi Arabian family presenting with mild AT features (Al-Muhaizea MA et al. Ann Hum Genet 2022 Jan;86(1):34-44). It was also detected in 1 of 7051 unselected breast cancer patients and 0 of 11241 female controls of Japanese ancestry (Momozawa Y et al. Nat Commun, 2018 Oct;9:4083). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
