This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 305 of the MEN1 protein (p.Gly305Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MEN1-related conditions. ClinVar contains an entry for this variant (Variation ID: 457344). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. The p.G305S variant (also known as c.913G>A) is located in coding exon 6 of the MEN1 gene. The glycine at codon 305 is replaced by serine, an amino acid with similar properties. This change occurs in the first base pair of coding exon 6. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
