The p.Y1400C variant (also known as c.4199A>G), located in coding exon 33 of the POLE gene, results from an A to G substitution at nucleotide position 4199. The tyrosine at codon 1400 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. -
