This variant changes 1 nucleotide in exon 5 of the BRCA2 gene, creating a premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. Splice site prediction tools suggest that this variant may not impact RNA splicing. To our knowledge, functional studies have not been performed for this variant. This variant has been reported in an individual affected with breast cancer (PMID: 27798748). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of BRCA2 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic. -
The p.S158* pathogenic mutation (also known as c.473C>G), located in coding exon 4 of the BRCA2 gene, results from a C to G substitution at nucleotide position 473. This changes the amino acid from a serine to a stop codon within coding exon 4. This alteration was reported in an indivdiual with a personal and family history of early onset breast cancer in a cohort of 190 Candadian women (Moran O et al. Breast Cancer Res. Treat., 2017 01;161:135-142). In addition to the information presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -

The substitution creates a nonsense variant, which changes a Serine to a premature stop codon (TCA>TGA), and is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. This variant, also known as BRCA2 701C>G using alternate nomenclature, has been reported in association with hereditary breast cancer (Moran 2016) and is considered pathogenic. -
