This variant is denoted BRCA2 c.5626G>T at the cDNA level and p.Glu1876Ter (E1876X) at the protein level. The substitution creates a nonsense variant, which changes a Glutamic Acid to a premature stop codon (GAA>TAA), and is predicted to cause loss of normal protein function through protein truncation. This variant, also known as 5857G>T or 5854G>T using alternate nomenclature, has been reported in a family with breast cancer (Phelan 1996) and is considered pathogenic.

The p.E1876* pathogenic mutation (also known as c.5626G>T), located in coding exon 10 of the BRCA2 gene, results from a G to T substitution at nucleotide position 5626. This changes the amino acid from a glutamic acid to a stop codon within coding exon 10. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
Hereditary breast ovarian cancer syndrome Pathogenic:1
This sequence change creates a premature translational stop signal (p.Glu1876*) in the BRCA2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in BRCA2 are known to be pathogenic (PMID: 20104584). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with BRCA2-related conditions. ClinVar contains an entry for this variant (Variation ID: 51894).
