The BRCA2 c.5897A>G (p.His1966Arg) variant has been reported in the published literature in individuals with breast and or ovarian cancer (PMID: 17972171 (2008), 18284688 (2008), 32438681 (2020), 34572941 (2021), 34178674 (2021), 35402282 (2022)) and described to be located in a region of the BRCA2 gene that is tolerant to missense sequence changes (PMID: 31911673 (2020)). In a large-scale breast cancer association study, the variant was observed in individuals with breast cancer as well reportedly healthy individuals (PMID: 33471991 (2021), see also see also LOVD (http://databases.lovd.nl/shared)). The frequency of this variant in the general population, 0.000071 (3/42136 chromosomes (Genome Aggregation Database, http://gnomad.broadinstitute.org)), is uninformative in the assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign. Based on the available information, we are unable to determine the clinical significance of this variant. -

Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.5e-05 in 1613942 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in BRCA2 causing Hereditary Breast And Ovarian Cancer Syndrome (3.5e-05 vs 0.00075), allowing no conclusion about variant significance. c.5897A>G has been reported in the literature in individuals affected with breast and/or ovarian cancer without strong evidence of causality (e.g. Lee_2008, Hondow_2011, Gorringe_2008, Santonocito_2020, Fanale_2021, Abdel-Razeq_2021, Patruno_2021, Abdel-Razeq_2022). These reports do not provide unequivocal conclusions about association of the variant with Hereditary Breast And Ovarian Cancer Syndrome.
