To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with breast cancer (PMID: 25682074). This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

The p.I2177T variant (also known as c.6530T>C), located in coding exon 10 of the BRCA2 gene, results from a T to C substitution at nucleotide position 6530. The isoleucine at codon 2177 is replaced by threonine, an amino acid with similar properties. This alteration was identified in an individual diagnosed with triple negative breast cancer (Wong-Brown MW et al. Breast Cancer Res Treat, 2015 Feb;150:71-80). This amino acid position is not well conserved in available vertebrate species, and threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
