This changes the amino acid from a tyrosine to a stop codon within coding exon 22. This alteration was identified in an individual with a personal and family history of breast and/or peritoneal cancer from a cohort of 1191 Chinese cancer index patients undergoing clinical evaluation and testing with multigene panels (Chan GHJ et al. Oncotarget, 2018 Jul;9:30649-30660). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay.
