The c.20519T>A (p.V6840D) alteration is located in exon 116 (coding exon 115) of the SYNE2 gene. This alteration results from a T to A substitution at nucleotide position 20519, causing the valine (V) at amino acid position 6840 to be replaced by an aspartic acid (D). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

This sequence change replaces valine with aspartic acid at codon 6840 of the SYNE2 protein (p.Val6840Asp). The valine residue is weakly conserved and there is a large physicochemical difference between valine and aspartic acid. This variant is present in population databases (rs758858068, ExAC 0.01%). This variant has not been reported in the literature in individuals with SYNE2-related disease.
