The c.763A>G (p.K255E) alteration is located in coding exon 4 of the SPG11 gene. This alteration results from an A to G substitution at nucleotide position 763, causing the lysine (K) at amino acid position 255 to be replaced by a glutamic acid (E). Based on data from the Genome Aggregation Database (gnomAD) database, the SPG11 c.763A>G alteration was observed in 0.01% (39/282828) of total alleles studied, with a frequency of 0.14% (34/24962) in the African subpopulation. The p.K255 amino acid is not conserved in available vertebrate species. The p.K255E alteration is predicted to be tolerated by in silico analysis. Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
