POLG (HGNC:9179): (DNA polymerase gamma, catalytic subunit) Mitochondrial DNA polymerase is heterotrimeric, consisting of a homodimer of accessory subunits plus a catalytic subunit. The protein encoded by this gene is the catalytic subunit of mitochondrial DNA polymerase. The encoded protein contains a polyglutamine tract near its N-terminus that may be polymorphic. Defects in this gene are a cause of progressive external ophthalmoplegia with mitochondrial DNA deletions 1 (PEOA1), sensory ataxic neuropathy dysarthria and ophthalmoparesis (SANDO), Alpers-Huttenlocher syndrome (AHS), and mitochondrial neurogastrointestinal encephalopathy syndrome (MNGIE). The missense variant c.3488T>A (p.Met1163Lys) in POLG gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Met1163Lys variant is novel (not in any individuals) in gnomAD exomes and 1000 Genomes. This variant has not been reported to the ClinVar database. The amino acid Met at position 1163 is changed to a Lys changing protein sequence and it might alter its composition and physico-chemical properties.The amino acid change p.Met1163Lys in POLG is predicted as conserved by GERP++ and PhyloP across 100 vertebrates.
