The c.3379C>G (p.L1127V) alteration is located in exon 26 (coding exon 25) of the MYH11 gene. This alteration results from a C to G substitution at nucleotide position 3379, causing the leucine (L) at amino acid position 1127 to be replaced by a valine (V). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
This missense variant replaces leucine with valine at codon 1134 of the MYH11 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function (internally defined REVEL score threshold 0.5 < inconclusive < 0.7, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with MYH11-related disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD).
