This variant has been reported in an individual affected with triple negative breast cancer in the literature (Lovejoy LA et al. 2018, ISSN : 2381-909X). This variant has been identified in 5/251484 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -

Variant summary: CDH1 c.1697T>C (p.Ile566Thr) results in a non-conservative amino acid change located in the Cadherin-like domain (IPR002126) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 1.1e-05 in 1613588 control chromosomes, predominantly at a frequency of 1.4e-05 within the Non-Finnish European subpopulation in the gnomAD database (gnomAD v.4.1.0). This frequency is not significantly higher than estimated for a pathogenic variant in CDH1 causing Breast Cancer (1.1e-05 vs 2.1e-05), allowing no conclusion about variant significance. c.1697T>C has been reported in the literature in at least an individual affected with endometrial and ovarian cancer (example: Na Kim_2022). These report(s) do not provide unequivocal conclusions about association of the variant with breast cancer. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
