This sequence change deletes 1 nucleotide from exon 34 of the NF1 mRNA (c.4640delA), causing a frameshift at codon 1547. This creates a premature translational stop signal (p.Lys1547Serfs*6) and is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in NF1 are known to be pathogenic. This particular variant has been reported in an individual affected with neurofibromatosis type 1 (NF1) (PMID: 21354044).
