The alanine at codon 1711 is replaced by glycine, an amino acid with similar properties. While this exact alteration has not been reported in the literature, a different alteration impacting the same codon, p.Ala1711Pro, was identified in a cohort of 378 individuals with suspected or clinically diagnosed neurofibromatosis type 1 (Xu W et al. Int. J. Mol. Med., 2014 Jul;34:53-60). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
