The p.S361* pathogenic mutation (also known as c.1082C>G), located in coding exon 9 of the BRCA1 gene, results from a C to G substitution at nucleotide position 1082. This changes the amino acid from a serine to a stop codon within coding exon 9. This alteration has been previously identified in Brazilian and European breast/ovarian cohorts (Esteves VF et al. Braz. J. Med. Biol. Res., 2009 May;42:453-7; Rebbeck TR et al. Hum. Mutat., 2018 05;39:593-620). Of note, this alteration is also designated C1201G in the published literature. In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
Hereditary breast ovarian cancer syndrome Pathogenic:1
Loss-of-function variants in BRCA1 are known to be pathogenic (PMID: 20104584). This variant has been observed in individuals and families affected with breast cancer and/or ovarian cancer (PMID: 19377795, 29446198). This variant is also known as C1201G in the literature. ClinVar contains an entry for this variant (Variation ID: 54121). This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Ser361*) in the BRCA1 gene. It is expected to result in an absent or disrupted protein product. For these reasons, this variant has been classified as Pathogenic. -
