The p.Q448K variant (also known as c.1342C>A), located in coding exon 10 of the SMAD4 gene, results from a C to A substitution at nucleotide position 1342. The glutamine at codon 448 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear. -
Juvenile polyposis syndrome Uncertain:1
This sequence change replaces glutamine with lysine at codon 448 of the SMAD4 protein (p.Gln448Lys). The glutamine residue is highly conserved and there is a small physicochemical difference between glutamine and lysine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SMAD4 protein function. This variant has not been reported in the literature in individuals with SMAD4-related conditions. This variant is not present in population databases (ExAC no frequency). -
