The p.L642W variant (also known as c.1925T>G), located in coding exon 13 of the LDLR gene, results from a T to G substitution at nucleotide position 1925. The leucine at codon 642 is replaced by tryptophan, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
