Variant classified as Uncertain Significance - Favor Benign. The Val6554Met vari ant in TTN has not been reported in the literature nor previously identified by our laboratory. This variant has been identified in 2/122 African American chrom osomes from a broad population by the 1000 Genomes project (dbSNP rs144032104). Computational analyses (biochemical amino acid properties, conservation, AlignGV GD, and SIFT) suggest that the variant may not impact the protein and orangutan carries a methionine (Met; this variant) at this position, raising the possibili ty that this change may be tolerated. However, this computational information is not predictive enough to rule out pathogenicity. Although this data supports th at the Val6554Met variant may be benign, additional studies are needed to fully assess its clinical significance. -
