Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), and loss-of-function variants in OTOF are known to be pathogenic (PMID: 18381613, 19250381, 22575033). This variant is present in population databases (rs111033455, gnomAD 0.04%). Disruption of this splice site has been observed in individual(s) with deafness (PMID: 22575033, 31379920, 34424407).
