Genes affected
OTOF (HGNC:8515): (otoferlin) Mutations in this gene are a cause of neurosensory nonsyndromic recessive deafness, DFNB9. The short form of the encoded protein has 3 C2 domains, a single carboxy-terminal transmembrane domain found also in the C. elegans spermatogenesis factor FER-1 and human dysferlin, while the long form has 6 C2 domains. The homology suggests that this protein may be involved in vesicle membrane fusion. Several transcript variants encoding multiple isoforms have been found for this gene. Variant classified as Uncertain Significance - Favor Benign. The Cys113Arg varia nt in OTOF has not been reported in individuals with hearing loss, but has been identified in 0.02% (2/8600) of European American chromosomes by the NHLBI Exome Sequencing Project (http://evs.gs.washington.edu/EVS/; dbSNP rs146615166). The amino acid (Cys) at position 113 is not conserved in mammals or evolutionary dis tant species, raising the possibility that a change at this position may be tole rated. Additional computational prediction tools suggest that the Cys113Arg vari ant may not impact the protein, though this information is not predictive enough to rule out pathogenicity. In summary, while the clinical significance of the C ys113Arg variant is uncertain, the conservation data suggest that it is more lik ely to be benign. -

The c.337T>C (p.C113R) alteration is located in exon 5 (coding exon 5) of the OTOF gene. This alteration results from a T to C substitution at nucleotide position 337, causing the cysteine (C) at amino acid position 113 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
