It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 90520). This pathogenic mutation has been identified in multiple HNPCC/Lynch syndrome families and one proband was reported to have a Lynch-associated tumor that demonstrated high microsatellite instability (Colombino M et al. Ann. Oncol., 2003 Oct;14:1530-6; Wolf B et al. Wien Klin Wochenschr, 2005 Apr;117:269-77; Colombino M et al. Eur. J. Cancer, 2005 May;41:1058-64; Ambry internal data). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). In silico splice site analysis predicts that this alteration will weaken the native splice donor site; however, direct evidence is insufficient at this time (Ambry internal data). In addition to the clinical data presented in the literature, alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
