This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of MSH6-related conditions (PMID: 24710284). ClinVar contains an entry for this variant (Variation ID: 483764). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is not expected to disrupt MSH6 protein function with a negative predictive value of 95%. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The p.M844T variant (also known as c.2531T>C), located in coding exon 4 of the MSH6 gene, results from a T to C substitution at nucleotide position 2531. The methionine at codon 844 is replaced by threonine, an amino acid with similar properties. This alteration was detected in 1/59 unrelated families from Singapore who met Amsterdam I or II criteria for Lynch syndrome, and it was classified as a variant of uncertain significance (Liu Y et al. PLoS ONE, 2014 Apr;9:e94170). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
