Genes affected
SYNJ1 (HGNC:11503): (synaptojanin 1) This gene encodes a phosphoinositide phosphatase that regulates levels of membrane phosphatidylinositol-4,5-bisphosphate. As such, expression of this enzyme may affect synaptic transmission and membrane trafficking. Multiple transcript variants encoding different isoforms have been found for this gene. The c.4603T>A (p.S1535T) alteration is located in exon 32 (coding exon 32) of the SYNJ1 gene. This alteration results from a T to A substitution at nucleotide position 4603, causing the serine (S) at amino acid position 1535 to be replaced by a threonine (T). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

This sequence change replaces serine, which is neutral and polar, with threonine, which is neutral and polar, at codon 1535 of the SYNJ1 protein (p.Ser1535Thr). This variant is present in population databases (rs138122167, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with SYNJ1-related conditions. ClinVar contains an entry for this variant (Variation ID: 478359). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
