MLH1 (HGNC:7127): (mutL homolog 1) The protein encoded by this gene can heterodimerize with mismatch repair endonuclease PMS2 to form MutL alpha, part of the DNA mismatch repair system. When MutL alpha is bound by MutS beta and some accessory proteins, the PMS2 subunit of MutL alpha introduces a single-strand break near DNA mismatches, providing an entry point for exonuclease degradation. The encoded protein is also involved in DNA damage signaling and can heterodimerize with DNA mismatch repair protein MLH3 to form MutL gamma, which is involved in meiosis. This gene was identified as a locus frequently mutated in hereditary nonpolyposis colon cancer (HNPCC). [provided by RefSeq, Aug 2017]

This variant is denoted MLH1 c.1897-3C>T or IVS16-3C>T and consists of a C>T nucleotide substitution at the -3 position of intron 16 of the MLH1 gene. Multiple in silico models predict this variant to either weaken the nearby natural splice acceptor site or increase use of a nearby cryptic splice acceptor site, possibly causing abnormal gene splicing. However, in the absence of RNA or functional studies, the actual effect of this variant is unknown. This variant has not, to our knowledge, been published in the literature as pathogenic or benign. The cytosine (C) nucleotide that is altered is not conserved. Based on currently available information, it is unclear whether MLH1 c.1897-3C>T is pathogenic or benign.
