In summary, this is a novel nonsense variant occurring in the final exon of the SCN11A gene. There is no indication that this variant causes disease, but the evidence is insufficient at this time to prove that conclusively. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have not been reported for this truncating variant and it is currently unknown if the last 34 amino acids of the SCN11A protein are critical for its function. This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with an SCN11A-related disease. This sequence change results in a premature translational stop signal in the last exon of the SCN11A mRNA at codon 1758 (p.Asp1758*). While this is not anticipated to result in nonsense mediated decay, it is expected to delete the last 34 amino acids of the SCN11A protein. -
