To the best of our knowledge, the variant has not been reported in the published literature. The frequency of this variant in the general population, 0.00024 (6/24952 chromosomes in African/African American subpopulation, http://gnomad.broadinstitute.org), is higher than would generally be expected for pathogenic variants in this gene. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is damaging. Based on the available information, we are unable to determine the clinical significance of this variant. -
In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; Observed with a second SDHA variant in a family with a suspected mitochondrial disorder; however, it is unknown whether the two variants were on the same (in cis) or opposite (in trans) alleles (PMID: 34732400); Observed in individuals with breast cancer in published literature (PMID: 33606809); This variant is associated with the following publications: (PMID: 34732400, 33606809) -
Dilated cardiomyopathy 1GG Uncertain:1

Pheochromocytoma/paraganglioma syndrome 5;C5700310:Mitochondrial complex II deficiency, nuclear type 1 Uncertain:1
This sequence change replaces arginine, which is basic and polar, with threonine, which is neutral and polar, at codon 97 of the SDHA protein (p.Arg97Thr). This variant is present in population databases (rs371274523, gnomAD 0.03%). This missense change has been observed in individual(s) with clinical features of autosomal recessive SDHA-related condition(s) (PMID: 34732400). ClinVar contains an entry for this variant (Variation ID: 539690). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on SDHA protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.R97T variant (also known as c.290G>C), located in coding exon 3 of the SDHA gene, results from a G to C substitution at nucleotide position 290. The arginine at codon 97 is replaced by threonine, an amino acid with similar properties. This alteration was detected in 1/224 unrelated Brazilian individuals with breast cancer (Sandoval RL et al. PLoS One, 2021 Feb;16:e0247363). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
SDHA-related disorder Uncertain:1
The SDHA c.290G>C variant is predicted to result in the amino acid substitution p.Arg97Thr. This variant was reported in an individual with breast cancer (Sandoval et al. 2021. PubMed ID: 33606809). This variant is reported in 0.024% of alleles in individuals of African descent in gnomAD (http://gnomad.broadinstitute.org/variant/5-224614-G-C), and it is interpreted as uncertain in ClinVar (https://www.ncbi.nlm.nih.gov/clinvar/variation/539690/). At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence. -
