SDHA (HGNC:10680): (succinate dehydrogenase complex flavoprotein subunit A) This gene encodes a major catalytic subunit of succinate-ubiquinone oxidoreductase, a complex of the mitochondrial respiratory chain. The complex is composed of four nuclear-encoded subunits and is localized in the mitochondrial inner membrane. Mutations in this gene have been associated with a form of mitochondrial respiratory chain deficiency known as Leigh Syndrome. A pseudogene has been identified on chromosome 3q29. The p.Y543* pathogenic mutation (also known as c.1629T>A), located in coding exon 12 of the SDHA gene, results from a T to A substitution at nucleotide position 1629. This changes the amino acid from a tyrosine to a stop codon within coding exon 12. Another alteration, c.1629T>G, that leads to the same premature stop codon (p.Y543*) was identified in a patient with esophagogastric adenocarcinoma through paired tumor-normal sequencing (Schrader KA et al. JAMA Oncol, 2016 Jan;2:104-11). In addition to the clinical data presented in the literature, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
