Computational analyses (bioch emical amino acid properties, conservation, AlignGVGD, PolyPhen2, and SIFT) sugg est that the Glu732Lys variant may not impact the protein, though this informati on is not predictive enough to rule out pathogenicity. Additional information is needed to fully assess the clinical significance of the Glu732Lys variant. -

This sequence change replaces glutamic acid with lysine at codon 732 of the LAMA4 protein (p.Glu732Lys). The glutamic acid residue is highly conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs727504935, ExAC 0.01%). This variant has not been reported in the literature in individuals with LAMA4-related conditions. ClinVar contains an entry for this variant (Variation ID: 179538). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
