This alteration results from a G to A substitution at nucleotide position 451, causing the glutamic acid (E) at amino acid position 151 to be replaced by a lysine (K). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 151 of the TREM2 protein (p.Glu151Lys). This variant is present in population databases (rs79011726, gnomAD 0.05%). This missense change has been observed in individual(s) with TREM2-related conditions (PMID: 25886450, 28620717, 32638105). ClinVar contains an entry for this variant (Variation ID: 1517663). Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The lysine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. Experimental studies have shown that this missense change does not substantially affect TREM2 function (PMID: 27589997).
