Four copies of this protein interact with one copy of the KCNE2 protein to form a functional potassium channel. Mutations in this gene can cause long QT syndrome type 2 (LQT2). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces arginine, which is basic and polar, with glutamine, which is neutral and polar, at codon 56 of the KCNH2 protein (p.Arg56Gln). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with long QT syndrome (PMID: 10973849, 11854117). ClinVar contains an entry for this variant (Variation ID: 67228). Experimental studies have shown that this missense change affects KCNH2 function (PMID: 10187793, 15475579, 20876384, 21536673, 25008322, 25809256, 25923442, 29725305, 30533098, 30988392, 31557540, 34716268).
