PTCH1 (HGNC:9585): (patched 1) This gene encodes a member of the patched family of proteins and a component of the hedgehog signaling pathway. Hedgehog signaling is important in embryonic development and tumorigenesis. The encoded protein is the receptor for the secreted hedgehog ligands, which include sonic hedgehog, indian hedgehog and desert hedgehog. Following binding by one of the hedgehog ligands, the encoded protein is trafficked away from the primary cilium, relieving inhibition of the G-protein-coupled receptor smoothened, which results in activation of downstream signaling. Mutations of this gene have been associated with basal cell nevus syndrome and holoprosencephaly. [provided by RefSeq, Aug 2017]

​The p.Y1009X pathogenic mutation (also known as c.3027C>A), located in coding exon 18 of the PTCH1 gene, results from a C to A substitution at nucleotide position 3027. This changes the amino acid from a tyrosine to a stop codon within coding exon 18. This mutation was identified in an individual diagnosed with nevoid basal cell carcinoma syndrome (Wicking, C et al. Am J Hum Genet. 1997 Jan;60(1):21-6). In addition to the clinical data presented in the literature, since premature stop codons are typically deleterious in nature, this alteration is interpreted as a disease-causing mutation (ACMG Recommendations for Standards for Interpretation and Reporting of Sequence Variations. Revision 2007. Genet Med. 2008;10:294). This mutation is also called C3015A in published literature.
