It is also involved in other processes including cell migration and adhesion, transcriptional activation, and apoptosis. In summary, this variant is a novel missense change with uncertain impact on mRNA splicing and protein function. It has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may alter RNA splicing, but this prediction has not been confirmed by published transcriptional studies. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0"). This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with an APC-related disease. This sequence change replaces asparagine with aspartic acid at codon 933 of the APC protein (p.Asn933Asp). The asparagine residue is highly conserved and there is a small physicochemical difference between asparagine and aspartic acid.

The p.N933D variant (also known as c.2797A>G), located in coding exon 15 of the APC gene, results from an A to G substitution at nucleotide position 2797. The asparagine at codon 933 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, in silico predictors for this gene do not accurately predict pathogenicity. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
