NM_000057.4:c.3500C>A
Variant summary
Our verdict is Uncertain significance. The variant received 3 ACMG points: 3P and 0B. PM2PP3
The NM_000057.4(BLM):c.3500C>A(p.Ala1167Glu) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.00000657 in 152,110 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. A1167G) has been classified as Uncertain significance.

Hereditary cancer-predisposing syndrome Uncertain:1
The p.A1167E variant (also known as c.3500C>A), located in coding exon 17 of the BLM gene, results from a C to A substitution at nucleotide position 3500. The alanine at codon 1167 is replaced by glutamic acid, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
