To our knowledge, functional studies have not been reported for this variant. This variant has been reported in multifactorial analyses with likelihood ratios for pathogenicity based on co-occurrence with a pathogenic variant and personal and family history (for 1 carrier) reaching a combined LR = 1.5020861552896 (PMID: 31131967, 31853058). This variant has been identified in 1/226418 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. -
The p.Q864L variant (also known as c.2591A>T), located in coding exon 10 of the BRCA2 gene, results from an A to T substitution at nucleotide position 2591. The glutamine at codon 864 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
