This missense variant replaces serine with leucine at codon 1404 of the BRCA2 protein. Computational prediction suggests that this variant may not impact protein structure and function (internally defined REVEL score threshold <= 0.5, PMID: 27666373). To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with ovarian cancer (PMID: 24504028) and in an individual affected with breast cancer (PMID: 33471991). This variant has been identified in 5/267258 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

The BRCA2 c.4211C>T; p.Ser1404Leu variant (rs41293489), is reported in the literature in a cohort of individuals affected with ovarian cancer, although its clinical significance in this study was uncertain (Cunningham 2014). This variant is found on five chromosomes (5/267258 alleles) in the Genome Aggregation Database. The serine at codon 1404 is moderately conserved, but computational analyses (SIFT: damaging, PolyPhen-2: benign) predict conflicting effects of this variant on protein structure/function.
