The MSH6 c.3586G>T; p.Glu1196Ter variant, to our knowledge, is not described in the medical literature or in gene-specific databases. It is also absent from general population databases (Exome Variant Server and Genome Aggregation Database), indicating it is not a common polymorphism. This variant induces an early termination codon and is predicted to result in a truncated protein or mRNA subject to nonsense-mediated decay. Additionally, several downstream truncating variants have been described in association with Lynch syndrome and are considered pathogenic (Berends 2002, Ferguson 2014, Terui 2013).
