Variant summary: KCNH2 c.889C>T (p.Pro297Ser) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The frequency data for this variant in gnomAD is considered unreliable, as metrics indicate poor data quality at this position. c.889C>T has been reported in the literature in individuals affected with Long QT Syndrome (Imboden_2006, Kapa_2009, Shimizu_2009, Giudicessi_2012, Winkel_2012, Itoh_2015, Gibbs_2018). These reports do not provide unequivocal conclusions about association of the variant with Arrhythmia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 30369311, 22949429, 17192539, 26669661, 19841300, 19926013, 22882672).
