The c.3491-3C>A intronic variant results from a C to A substitution 3 nucleotides upstream from coding exon 32 in the MYBPC3 gene. This nucleotide position is highly conserved in available vertebrate species. Another alteration affecting the same position, c.3491-3C>G, has been reported in association with hypertrophic cardiomyopathy (HCM) (Lopes LR et al. Heart, 2015 Feb;101:294-301). The BDGP splice prediction software does not produce a reliable prediction for the nearby native splice acceptor site. The ESEfinder splice prediction software predicts a weakening in the native splice acceptor site efficiency. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
