The c.214G>A (p.E72K) alteration is located in exon 3 (coding exon 2) of the PHKG2 gene. This alteration results from a G to A substitution at nucleotide position 214, causing the glutamic acid (E) at amino acid position 72 to be replaced by a lysine (K). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear.

This sequence change replaces glutamic acid with lysine at codon 72 of the PHKG2 protein (p.Glu72Lys). The glutamic acid residue is moderately conserved and there is a small physicochemical difference between glutamic acid and lysine. This variant is present in population databases (rs753644625, ExAC 0.009%). This variant has not been reported in the literature in individuals affected with PHKG2-related conditions.
