Variant summary
Our verdict is Uncertain significance. The variant received 2 ACMG points: 3P and 1B. PM1PP2BP6
The NM_000388.4(CASR):c.1826C>T(p.Thr609Met) variant causes a missense change. The variant allele was found at a frequency of 0.00000496 in 1,613,954 control chromosomes in the GnomAD database, with no homozygous occurrence. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. T609R) has been classified as Uncertain significance.

The p.T609M variant (also known as c.1826C>T), located in coding exon 6 of the CASR gene, results from a C to T substitution at nucleotide position 1826. The threonine at codon 609 is replaced by methionine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
