This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with hypophosphatasia (PMID: 15300736, 27312557, 28127875, 32160374). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. This variant is also known as Gly95Ser. ClinVar contains an entry for this variant (Variation ID: 549969). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt ALPL protein function with a positive predictive value of 95%. Variant summary: ALPL c.334G>A (p.Gly112Ser) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251008 control chromosomes. c.334G>A has been reported in the literature in multiple individuals affected with Hypophosphatasia (Witters_2004, Park_2016, Tenorio_2017, Del Angel_2020, Mornet_2020). These data indicate that the variant is very likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 32160374, 38702915, 32973344, 27312557, 28127875, 15300736).
