This missense change has been observed in individual(s) with Charcot-Marie-Tooth disease (PMID: 10463363). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces aspartic acid, which is acidic and polar, with asparagine, which is neutral and polar, at codon 128 of the MPZ protein (p.Asp128Asn). ClinVar contains an entry for this variant (Variation ID: 242747). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts the p.Asp128 amino acid residue in MPZ. Other variant(s) that disrupt this residue have been observed in individuals with MPZ-related conditions (PMID: 9888385, 18636082), which suggests that this may be a clinically significant amino acid residue.
