In the published literature, the variant has been reported in an individual with tuberous sclerosis complex (TSC) (PMID: 32917966 (2021)). It has also been reported to have no deleterious effect on TSC2 target kinase activity (PMID: 21309039 (2011)), however additional studies are required to determine the global effect of this variant on TSC2 protein function. The frequency of this variant in the general population, 0.000065 (2/30608 chromosomes, http://gnomad.broadinstitute.org), is uninformative in assessment of its pathogenicity. Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is benign.
