The Q220X nonsense variant in the TSC2 gene has been reported previously in association with tuberous sclerosis complex (TSC) (TSC2 LOVD). This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q220X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Therefore, the presence of Q220X is consistent with the diagnosis of TSC in this individual. -
The TSC2 c.658C>T p.Gln220Ter variant (rs45517119), is reported in databases in individuals affected with tuberous sclerosis (Stenson 2012, LOVD). The TSC2 c.658C>T p.Gln220Ter variant is also reported in ClinVar (Variation ID: 49411). This variant is absent from the Genome Aggregation Database, indicating it is not a common polymorphism.
