This variant has been observed in individual(s) with bilateral multifocal renal oncocytomas and cysts; however, this individual had a co-occurring translocation that may be responsible for the phenotype (PMID: 9523203). ClinVar contains an entry for this variant (Variation ID: 809420). The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This sequence change replaces proline with arginine at codon 40 of the VHL protein (p.Pro40Arg). The proline residue is weakly conserved and there is a moderate physicochemical difference between proline and arginine. -
