NM_001999.4:c.7655G>A
Variant summary
Our verdict is Uncertain significance. The variant received 5 ACMG points: 6P and 1B. PM2PP3_StrongBP6
The NM_001999.4(FBN2):c.7655G>A(p.Gly2552Glu) variant causes a missense change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.0000031 in 1,613,418 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a pathogenic outcome for this variant. 13/22 in silico tools predict a damaging outcome for this variant. Variant has been reported in ClinVar as Conflicting classifications of pathogenicity (no stars). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. G2552A) has been classified as Uncertain significance.

Familial thoracic aortic aneurysm and aortic dissection Uncertain:1
The p.G2552E variant (also known as c.7655G>A), located in coding exon 60 of the FBN2 gene, results from a G to A substitution at nucleotide position 7655. The glycine at codon 2552 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
not provided Uncertain:1
Not observed at significant frequency in large population cohorts (gnomAD); In silico analysis supports that this missense variant has a deleterious effect on protein structure/function; Has not been previously published as pathogenic or benign to our knowledge
Congenital contractural arachnodactyly Benign:1
